Upload the MKV and open the options. Frame Extraction has two modes: Single Frame saves one TIFF from the second you enter (for example 1, 5 or 10.5), and All Frames saves every frame of the video as a TIFF image. There is no every-Nth-frame or one-per-second mode.

HDR footage is not tone-mapped: each frame is converted from the MKV as it is decoded, so a frame from an HDR MKV can look flat or washed out as a TIFF. Standard (SDR) video keeps its colours.

It depends on the size and length of the file. Small documents and images are usually ready quickly. Long videos take the longest, because most video conversions re-encode every frame.

One at a time. Single Frame takes one time in seconds (for example 83 for 1:23) and saves that frame as a TIFF. There is no field for a list of times, so run the conversion again for each extra timestamp.

No. The converter has no scene detection and no one-per-second mode: it saves either a single frame at the second you enter or every frame. For one image per scene, extract single frames at the times where the scenes change.
